Abstract





The solution for staining rinses in peripheral blood smear preparations (SADT) is very important especially with regard to pH. A rinse solution that is too acidic can cause decolorization of the SADT. WHO recommends rinsing solutions using phosphate buffers, especially pH 7.2. This study aims to determine the picture of erythrocytes using 5% Giemsa staining and Wright by rinsing using tap water and phosphate buffer pH 7.2. The method used is a descriptive method with a sample in the form of EDTA blood from 2 (two) Thalassemia patients and 5 (five) people from D III Medical Laboratory Technology students, Faculty of Health Sciences, University of Muhammadiyah Palangkaraya. Each of the samples was then given 4 (four) treatments, namely Giemsa staining with tap water rinsing and phosphate buffer pH 7.2 and Wright staining with tap water rinsing and phosphate buffer pH 7.2. A total of 28 slides of edge blood swab preparations were obtained which were then microscopically examined to determine the picture of erythrocytes. In both normal and pathological samples, erythrocytes appeared pink with a clear centralpallor on Giemsa staining of 5% and more contrast on Wright. The background of the observation is lighter when rinsed with tap water, while it is cleaner although slightly dark when rinsed with a pH phosphate buffer of 7.2. Overall, Giemsa's 5% and Wright stains were both usable for erythrocyte observations, and erythrocyte stains were not significantly affected by tap water flushing despite their acidic pH (5.0).
